ESTUDOS DE BIOEQUIVALÊNCIA PARA FÁRMACOS QUE APRESENTAM ESTEREOISOMERISMO
Abstract
A maioria dos fármacos que apresentam esteroisomerismo é comercializada na forma de racemato (mistura deenantiômeros). Isto se deve, inicialmente, à falta de tecnologia apropriada (de síntese ou analítica) e à razão econômica, já que é menos dispendioso produzir racematos do que enantiômeros puros (Welage e cols., 2001). Atualmente, o avanço tecnológico tornou mais acessível, tanto a produção e o controle de qualidade, quanto à determinação de concentrações plasmáticas dos enantiômeros individuais. Desta forma, há necessidade de se avaliar o impacto do esteroisomerismo nos estudos de bioequivalência, empregados, principalmente para assegurar a intercambialidade entre medicamentos genéricos, ou similares, e de referência(Anvisa, RE nº 896). Em função do exposto, pretendemos analisar criticamente as principais legislações quanto às suas orientações em relação ao planejamento e execução de estudos de bioequivalência para fármacos que apresentam esteroisomerismo. Para isto, é necessário, primeiramente, destacar qual pode ser a influência do estereoisomerismo na farmacodinâmica e farmacocinética do fármaco.Downloads
